維生素D受體基因FokⅠ位點多態性與漢族人群中重度牙周炎易感性的關系

摘要:目的:探討維生素D 受體(VDR) 基因FokⅠ位點多態性與漢族人群中重度慢性牙周炎 (CP) 易感性的關系。方法:搜集漢族中重度牙周炎病人71例,并以50例牙周健康者作為對照組,均采外周靜 脈血提取基因組DNA,采用聚合酶鏈反應- 限制性片段長度多態性( PCR - RFLP)方法測定FokⅠ基因型,并分 析組間基因型和等位基因頻率的差異。結果: CP組的基因型分布情況為: FF (24, 33. 8% ) , Ff (38, 53. 5% ) 和ff (9, 12. 7% ) ,對照組為: FF ( 9, 18% ) , Ff ( 26, 52% ) 及ff ( 15, 30% ) , 組間比較有顯著性差異(χ2 = 7. 14, P = 0. 028) 。CP組等位基因頻率為F (60. 6% ) , f (39. 4% ) ,對照組為F (44% ) , f (56% ) ,組間比較有顯著性差 異(χ2 = 6. 48, P = 0. 011) 。FF基因型相對于( Ff + ff)的OR = 2. 33 (95%CI = 0. 97 - 5. 57) ; F等位基因相對于f 的OR = 1. 95 (95%CI = 1. 16 - 3. 28) 。結論:VDR基因FokⅠ位點多態性與漢族人中重度牙周炎易感性可能存 在一定的聯系。

Association of vitam in D receptor gene fokⅠpolymorphism with moderate to severe chron ic per iodontitis of han nationality

ABSTRACT:A IM : To investigate the association of vitamin D recep tor gene FokⅠ polymorphism with moderate to severe chronic periodontitis of Han Nationality. METHODS: A restriction fragment length polymorphism (RFLP) for FokⅠ of the VDR gene was analyzed by polymerase chain reaction ( PCR) , followed by restriction digestion with FokⅠ and gel electrophoresis. The genotypes of 71 moderate severe cases of periodontitis and 50 controlswith no periodontitis were analyzed. RESULTS: The genotype distribution in the patient group were 24 ( 33. 8% ) , 38 (53. 5% ) and 9 (12. 7% ) and in the control group were 9 (18% ) , 26 (52% ) and 15 (30% ) for FF, Ff and ff respectively. Allele frequencies were 60. 6% and 39. 4% for F and f in the patient group; 44% and 56% in the control group. Chi square analysis indicated that the distribution of the genotypes and alleles between these two group swere sig nificantly different ( P = 0. 028, P = 0. 011, respectively). The F allele seemed to increase suscep tibility to chronic periodontitis (OR = 1. 95, 95%CI = 1. 16 - 3. 28). CONCLUS ION: It indicated that okⅠpolymorphism of VDR gene was associated with chronic periodontitis of Han nationality.
